Virus resistance research in potato is crucial, especially in African countries where most seed tubers are infected with viruses like Potato virus Y (PVY) and Potato leaf roll virus (PLRV), leading to significant yield losses. In contrast, industrial nations experience minimal virus issues due to effective seed certification. However, PVY and PLRV remain major global threats due to their rapid evolution and potential to overcome resistances. Recently, common PVY strains have been increasingly replaced by virulent recombinant strains, which can cause serious diseases like potato tuber necrotic ring spot disease (PTNRD). This situation necessitates a deeper understanding of PVY and PLRV epidemiology and the introduction of new resistance genes in breeding programs tailored to local climates. Reliable virus detection methods are essential for these efforts. This thesis presents the development and application of reverse transcription real-time PCR (RT-qPCR) assays for quantifying PVY and PLRV, which can significantly aid potato research. The RT-qPCR assay for PVY is highly sensitive, enabling direct testing of freshly harvested tubers during seed certification. It also evaluates the resistance levels of various Solanum species. Additionally, RT-qPCR assesses PLRV virulence, although a direct correlation with titer was not established.
